C2C training

Having just completed the C2C the wrong way round in 2 days on my MTB as part of a charity event I now want to challenge myself to doing the C2C in one day on my road bike for next year.
Things I have learned this year, some of the hills heading into Alston killed me, Winlatter pass the wrong way is nuts, I was tentative on some of the downhill sections (and still hit 47mph...big hills) and overall wasn't too bad considering I have only been cycling for 6-7 months. I think saddle time was around 15hours and total time out was 17 hours over 2 days. I now have a computer as well as a HR monitor so that should aid training.
Now the question is what would you recommend I do to build strength and endurance of my legs as well as build confidence on the quick bits?

Probably should add a caveat.........increase your weekly mileage or riding hours steadily - a week-on-week increase of no more than 10% maximum. Don't want to be responsible for you getting tendonitis or something equally unpleasant.
If you keep plugging away at it (and keep riding through the winter), gradually increasing your mileage, you'll find a 140-mile hilly ride fairly straight-forward come next summer.0
