Canyon endurace vs Giant Defy vs wildcard

I'm looking at my next cycle to work scheme bike, so up to £1k.
My main priority is comfort as I get lower back pain fairly often (professional fittings have helped only a bit).
I'm currently on a Giant defy (from 2015, the last model or so before they went all disk) and am keen to get input on what I can expect to find different if I switched to a Canyon endurace AL 6.0. Or, if you have a wildcard idea, what would that be? Ideally non-disc but I'm not fussed (as I have some campag zondas I would prefer to re-use rather than sell).
