Hi guys, a few questions about my bike (http://www.giant-bicycles.com/en-au/bikes/print-model-pdf/12008/57410/)

1. Can I put a 11-32t 105 5800 cassette on it? If not what is the best way to get a 32t cassette on this?
2. Can I put 25 or 28mm tyres on it?

Thanks in advance

The bike you link to has 10-speed 105 fitted so no you would not be able to fit an 11-speed 105 5800 12-32 cassette without changing the shifters and lots of other bits.

However there is a solution - you could fit a Shimano SLX HG81-10, 11-32 cassette that will be perfectly compatible with your 10-speed shifters

One problem might be the rear derailleur. If you currently have the short cage (SS) version then you may need to buy the medium cage (GS) version to cope with the 32 sprocket. You will also probably need to lengthen your chain.

Not sure about the tyre question - could be a case of trial and error to see what fits. Sopme tyres size differently depending on the wheel rim they are fitted to.

You would need to change your existing 10sp 105 RD-5701 SS to a 105 RD-5701 GS which will work with a 10sp 11-32T cassette, either the SLX HG81-10 mentioned above or a CS-HG500-10. You would also need a longer chain.

Giants recommendation for your model Defy with the standard P-R2 wheels (622x14mm) is 23c or 25c tyres, you may be able to use 28c tyres, you would need to check the frame/fork clearances, and to the underside of the brake calipers.
