Converting to tubeless after puncture

I will be converting to tubeless tomorrow after suffering a puncture on my rear tyre. This may be a naive question but with the tyre being punctured will I be able it convert it to tubeless or do I need to be converting a tyre that hasn't yet been punctured in order for it to be successful? Surely the sealant will render the small puncture with a few rotations?

    The sealent should sort it out. You'll possibly even see more holes that didn't puncture the tube, and probably a bit of oozing out the sidewalls, depending on the tyres.
