I'm looking to take advantage of the current sales on a new set of tyres to replace my stock Giant defy but I'm undecided on 25's or 28's, I looking for a faster rolling and more comfortable tyre, I'm pretty much set on the Continental GP 4000S II but scratching my head on size.

You won't go far wrong with the 25mm gp4000s, these are slightly oversized any way, and give a very comfortable ride

But if your bike has plenty of clearance for 28s then why wouldn't you?

Being a Defy disc bike I'm sure there's clearance for 30's so no problem fitting 28's.

Very hilly in my area so would 28's be ok for climbs ?

Mine come out at 27.6mm on DT Swiss R23 rims.
Rose Xeon CDX 3100, Ultegra Di2 disc (nice weather)
Ribble Gran Fondo, Campagnolo Centaur (winter bike)
    Van Raam 'O' Pair
Land Rover (really nasty weather  )

^ sounds about right, 25mm GP4000s ii come out at pretty much exactly the same size as 28mm 4 Seasons in my experience. They are massive!

So be careful with clearances. I use 28mm in most of my bikes, but I think I would struggle a little with 28mm GP4000s ii
