Probably a silly question to most people , but if want to clean or take out pads do I have to remove the wheel first or can it be done with wheels left on ?

as per same question you asked in the other thread, answer is no, wheel can stay in, obvious if you think about it really.

But again it does depends on the brakes.

Why is it an issue one way or the other? Wheels take approx 1.2 seconds to remove.

ps 99.874% of all brake issues are caused by messing around with the brakes.
Just rinse with a bit of water in situ if muddy, otherwise leave them alone. Don't take them out.
