Does the cable need to be loosened off to set the LH limits on the front derailleur?

I would say maybe. If depends on what is happening.

I put a new drivechain in my old Scott (cassette, bb, crank, chain etc) On the first ride out I changed up to the big ring on the front and the chain went off the other side.

Knew this had something to do with the H limit. Turning the screw didnt seem to alter the position of the cage. This is what made me think 'do I need to loosen the cable' ?

    You should not have to.

Just turn the H limit 1/4 of a turn then test by chaning up on to the biggest ring.

Repeat until the cage stops trying to push the chain over the biggest ring.

IIRC, doing up the screw limits the throw off.

You don't have to as moving it inwards releases some tension, but too much and the cable will be a bit slack, so I loosen the cables for any adjustment - only takes a second anyway.
Set the limits, then fit the cable, then fine tune with the adjusters.

Except the cable will be pulling it hard into the H-stop.......the indexing clearly needs sorting as well!
