It's a great trip. I'd take three days and be leisurely about it. I don't know how you are getting out there but If you are heading out that way from Melbourne, one nice possibility is to ride down the Mornington Peninsula, on the long curve of Port Philip Bay south of the city, and drop down to Portsea - a very pretty ride - then cross on the short ferry to Queenstown and set off along the coast from there.
It is far, far better than riding out along the main highway to Geelong.
(I used to live in Melbourne, by the way)
There is plenty of accommodation - of all kinds and grades - along the route, and the scenery is stunning. Wandering down to Cape Otway is a nice detour.
I'd avoid the weekend, if possible, and go mid week as the traffic will be lighter.0 -

Other than that I am certain bike hire is possible, no specifics. Living there as I used to do, I had my own bike. I lived in Elsternwick, on in the city's inner south, and occasionally when the thought of riding into the city for another day at The Age newspaper, where I used to work, seemed too tedious, I used to take a 'wrong turn' on the way in and instead ride down the Mornington Peninsula, to the ferry, and then swing back home again the long way, via Geelong, a hundred-mile loop. A lot more fun than a day at the office!0
