Just Entered the London - Cambridge But no Road Bike!

I have just entered myself into the London - Cambridge Cycle ride but the only bike I have is my Cove Hustler, not sure this will be the best bike for the job so I am contemplating buying a Road Bike, so what should I buy and how do I know what size I should buy? Is sizing the same as MTB's?
My Budget will be Max £500 prefer £400
Sizing Info - I'm about 6'1
Prefer a new bike but would consider 2nd Hand.
