Flat spot on tyre?

I have noticed whilst on road going to a meeting point that above a certain speed my pads start to catch on my disc.
Today I carefully looked over my bike and there is a flat spot on my tyre. I prooved this by using a zip tie that just touched the top of the tyre. As the valve area passed the zip tie then the zip tie no longer touched the tyre, maybe by a mm or less.
I have removed and re-seated the inner tube which has made no difference.
So is this anything to worry about?
If so is it likely to be a faulty tyre, or could it be a dinged wheel? Not sure how to trouble shoot it any further at the moment so looking for assistance!

Are you serious?
A MTB tyre is never going to be perfectly round, and will probably seat slightly differently every time its removed and refitted.
Take your cable tie off and go ride the thing.0
