Dales Cycles or Alpine Bikes might hire you a road bike.
As for a 100km loop you could do Glasgow- Lennoxtown-over the Crow-down to the Carron Valley resevoir-over the Tak me Doon to Kilsyth then back to Glasgow. Probably just over a 100km

Alpine Bikes at the Glasgow Outdoor Experience definitely hire bikes, but I'm not sure if it's just MTBs. Give them a ring.

As for a 100km loop, try this one

It's quite a hilly route, but it can easily be shortened or increased depending on your fitness.
From the top of the Tak ma Doon you get a great view of central Scotland.

If you get the train out of Glasgow, it would avoid the busiest roads at the start of the loop.

All of these shops are reachable from Glasgow central some closer than others.

Arranandy's route over the Crow, along the valley and then over the Tak Me Doon is spot on but if you want to hit 100km's then when you get to the bottom of the Tak Me Doon then you would need to about turn and go back over the route you have just ridden which is a great route to do as the Tak is best approached from the Kilsyth side anyway.

If you go from Glasgow town centre-Torrance-Lennoxtown-Crow-Carron Valley-Carron bridge-Kilsyth then go back to Carron Bridge-along the valley-over the Crow and back to Glasgow you will hit 100km with approx 1600mtrs of climbing. Not sure about your level of riding but this would not be an easy ride so if you fancy it take your time and watch out on the Tak me Doon as there are some really steep bits and a small stream runs across the road on the descent from Kilsyth to carron Bridge which could easily catch you out.
