Fitting a new hydraulic brake hose

Right, I have a new hose and have removed the banjo, olive, etc from each end of the old one.
I've put them on the new hose and bolted everything back together.
Problem is, now I try and fill the system from the little hole in the lever cylinder, nothing will go (I am getting back-pressure on the syringe.
I've opened the bleeder on the caliper and pumped the lever, but it still won't take any fluid from the syringe.
I am about at the end of my patience - I used to be a bike mechanic and can strip and re-build any bike, but this was before hydraulic brakes!
I was thinking that the hose could be blocked? could it be anything else?

have you tried pumping the fluid in from the caliper by the syringe instead, do this till the resivoir is full to the brim, tap the lines a bit, give the lever a pump a couple of times (making sure you have a spacer in the pistons if you have took the pads out and done it properly, retighten bleed nipple, top up reserviour again, and reseal, job done.0
