Can the R5 frame set be fitted with standard mechanical Ultegra or Dura-ace?

Yes.

I think you may have to play around with the cable routing if you want to go Di2, but mechanical works fine.

Probably been covered on other threads, but you will of course need an adaptor for the bottom bracket.
