I've just changed my Sella Italia SLR to a Fizik Arione, now the Fizik is 30mm longer than the Sella Italia and as my set up was correct for the SLR do I use the same measurements from the nose to handlebar distance or take it from the rear of saddle? Hope I've explained myself

You want to measure to the part you sit on - the nose doesn't matter so that would make this one way to far back if you measured to there.
It can be a bit hard to tell so I would just look at them and look where you think you would be sitting and then put it there. You'll probably need to fiddle with it once you've got it on there.I'm left handed, if that matters.0 -
Adjusting seats is really subjective. My technique is to plonk it in the middle and see how it feels when climbing and just rolling along. Then keep stopping and make tiny adjustments back or forward depending if you feel too far back or forward and then keep going and seeing if it feels any better. You'll find you're doing lots of stopping and starting (I think I still adjusted my saddle ~5 times when I moved the seat to my new bike after I had had it for years), so take an allen key with you!
Also, the Arione does take some time for the wingflex to do its thing (The Arione was the seat I moved to my new bike, it's a great saddle!). So it could take you a few weeks to a month to know if you hate it or love it. I have mine angled down a fraction (as the nose does curve up a little).0
